Description: Cleavage Under Targets and Tagmentation (CUT&Tag.)

Technical Requirements/Tasks:
1. Contractor must be able to discuss the project and provide detailed protocols and guidance on preparing samples for shipping.
2. Contractor must be able to assess sample quality before starting the CUT&Tag assay. This includes, at minimum, a nuclear stain to assess for cell viability. The assay will be performed if the sample has at least 100,000 cells at >70% viability.
3. Contractor must provide all reagents and controls to perform the H3K27Ac assay (antibody required with reactivity to bovine chromatin) and control reactions. Batch controls with established cell line will be performed in parallel to ensure assay performance.
4. Contractor must perform Illumina sequencing of DNA isolated from the CUT&Tag assay and process sequencing data through respective pipeline, which includes sequence alignment to the bovine genome, read normalization, and peak calling.
5. Contractor must readily discuss results and provide a data report with all the raw sequencing data files and include genome browser views.
